The cell density of the diatom Thalassiosira genus, which is dominant in Tokyo Bay, was determined by light microscopy, an isolated culture was created, the species was identified by electron microscopy, and the 18S rDNA sequence was determined. did. Based on the sequence information, we compared the results obtained by DNA metabarcoding and found that Thalassiosira binata is overwhelmingly dominant among phytoplankton in the summer. Although the existence of the same species has been known for some time, optical microscopy alone cannot reveal the detailed morphology, and DNA metabarcoding does not allow correct genetic information to be found in the DNA database, so neither method can accurately identify the species. However, we were able to clarify this by combining isolation culture and both methods.
